    Efficacy of a curcumin extract (Curcugen™) on gastrointestinal symptoms and intestinal microbiota in adults with self-reported digestive complaints: a randomised, double-blind, placebo-controlled study

    Background There is preliminary evidence to suggest curcumin can alleviate digestive symptoms in adults with self-reported digestive complaints and irritable bowel syndrome. However, in all these trials, curcumin was used as a component of a multi-herbal combination and there were consistent concerns associated with risk of bias in most studies. The goal of this study was to investigate the effects of a curcumin extract (Curcugen™) on gastrointestinal symptoms, mood, and overall quality of life in adults presenting with self-reported digestive complaints. Moreover, to determine the potential therapeutic mechanisms of action associated with curcumin, its effects on intestinal microbiota and small intestinal bowel overgrowth (SIBO) were examined. Methods In this 8-week, parallel-group, double-blind, randomised controlled trial, 79 adults with self-reported digestive complaints were recruited and randomised to receive either a placebo or 500 mg of the curcumin extract, Curcugen™. Outcome measures included the Gastrointestinal Symptom Rating Scale (GSRS), intestinal microbial profile (16S rRNA), Depression, Anxiety, and Stress Scale – 21 (DASS-21), Short Form-36 (SF-36), and SIBO breath test. Results Based on self-report data collected from 77 participants, curcumin was associated with a significantly greater reduction in the GSRS total score compared to the placebo. There was also a greater reduction in the DASS-21 anxiety score. No other significant between-group changes in self-report data were identified. An examination of changes in the intestinal microbial profile and SIBO test revealed curcumin had no significant effect on these parameters. Curcumin was well-tolerated with no significant adverse events. Conclusions The curcumin extract, Curcugen™, administered for 8 weeks at a dose of 500 mg once daily was associated with greater improvements in digestive complaints and anxiety levels in adults with self-reported digestive complaints. Compared to the placebo, there were no significant changes in intestinal microbiota or SIBO; however, further research using larger samples and testing methods that allow more detailed microbial analyses will be important. An investigation into other potential mechanisms associated with curcumin’s gastrointestinal-relieving effects will also be important such as examining its influence on the intestinal barrier function, inflammation, neurotransmitter activity, and visceral sensitivity

    CAPTCHaStar! A novel CAPTCHA based on interactive shape discovery

    Over the last years, most websites on which users can register (e.g., email providers and social networks) adopted CAPTCHAs (Completely Automated Public Turing test to tell Computers and Humans Apart) as a countermeasure against automated attacks. The battle of wits between designers and attackers of CAPTCHAs led to current ones being annoying and hard to solve for users, while still being vulnerable to automated attacks. In this paper, we propose CAPTCHaStar, a new image-based CAPTCHA that relies on user interaction. This novel CAPTCHA leverages the innate human ability to recognize shapes in a confused environment. We assess the effectiveness of our proposal for the two key aspects for CAPTCHAs, i.e., usability, and resiliency to automated attacks. In particular, we evaluated the usability, carrying out a thorough user study, and we tested the resiliency of our proposal against several types of automated attacks: traditional ones; designed ad-hoc for our proposal; and based on machine learning. Compared to the state of the art, our proposal is more user friendly (e.g., only some 35% of the users prefer current solutions, such as text-based CAPTCHAs) and more resilient to automated attacks.Comment: 15 page

    Blue biotechnology: oil bioremediation using hydrocarbon-degrading bacteria immobilized on biodegradable membranes

    A novel bioremediation system to clean up oil contaminated water was developed combining hydrocarbon (HC) degrading bacteria immobilized and polylactic acid (PLA) or polycaprolactone (PCL) membranes prepared by electrospinning. The bioremediation efficiency was tested on crude oil using highly performant HC degrading bacterial strains isolated from marine and soil environments. The membrane morphology, the microbial adhesion and proliferation were evaluated using scanning electron microscopy (SEM). The SEM analysis highlighted that the fibers of the electrospun mats were in nanoscale with a similar diameter size distribution. The electrospun membranes exhibited high oil absorption capacity (q): approximately q = 40 g/g for PLA and q = 20 g/g for PCL. The bacterial strains were able to attach to the PLA and PCL membranes after 48h, reaching high proliferation and biofilm formation within the whole structure in 5 days. The biodegradation efficiency of the bacteria-membrane systems was tested by GC-FID analysis and compared with planktonic cells after 5 and 10 days incubation. The bacterial immobilization is a promoting factor for biodegradation and a new tool to be developed for bioremediation of aquatic systems

    Blue biotechnology: enhancement of bioremediation using bacterial biofilms on biodegradable scaffolds

    Petroleum hydrocarbons are still the most threatening environmental pollutants. A promising non invasive and low-cost technology for the treatment of contaminated sites is based on bioremediation by biodegrading microorganism endowed with catabolic ability towards oil and derivatives. New methods are needed to enhance and optimize natural biodegradation, such as the immobilization of hydrocarbons degraders in many types of supports. We developed a scaffold-bacteria bioremediation system to clean up oil contamination based on degradable 3D scaffolds. The polycaprolactone component is biodegradable, produced in the melt, i.e. at low cost and without the use of toxic solvents. The biofilm is made of highly performing HC-degrading bacteria such as the marine hydrocarbonoclastic bacteria (HCB) (1) or solid n-alkane degrading Actinobacteria (2, 3). The bacterial biofilm is observed within the whole structure of scaffold using scanning electron microscopy. The bioremediation efficiency of such systems was tested on crude oil by GC-FID analysis and compared whit planktonic cells. The biofilms formation was a promoting factor for biodegradation showing hydrocarbon removal up to 70% and 15% more in respect to the planktonic cells. Increasing availability of the contaminants and a better interaction between the hydrophobic substrate and the bacterial cells resulted in developing the degradation rate.     Evidence of destabilization of the human thymidylate synthase (hTS) dimeric structure induced by the interface mutation Q62R

    In human cells, thymidylate synthase (TS) provides the only source of 2\u2019-deoxythymidyne-5\u2019-monophosphate (dTMP), which is required for DNA biosynthesis. Because of its pivotal role, human TS (hTS) represents a validated target for anticancer chemotherapy. Nonetheless, the efficacy of drugs blocking the hTS active site has limitations due to the onset of resistance in cancer cells, requiring the identification of new strategies to effectively inhibit this enzyme. Human TS works as an obligate homodimer, making the inter-subunit interface an attractive targetable area. Here, we report the design and investigation of a new hTS variant, in which Gln62, located at the dimer interface, has been replaced by arginine in order to destabilize the enzyme quaternary assembly. The hTS Q62R variant has been characterized though kinetic assay, thermal denaturation analysis and X-ray crystallography. Our results provide evidence that hTS Q62R has a reduced melting temperature. The effective destabilization of the TS quaternary structure is also confirmed by structural analysis, showing that the introduced mutation induces a slight aperture of the hTS dimer. The generation of hTS variants having a more accessible interface area can facilitate the screening of interface-targeting molecules, providing key information for the rational design of innovative hTS interface inhibitors

    Effects of a Bacopa monnieri extract (Bacognize®) on stress, fatigue, quality of life and sleep in adults with self-reported poor sleep: A randomised, double-blind, placebo-controlled study

    In this 28-day, randomised, double-blind, placebo-controlled trial, 100 adults with self-reported poor sleep received either a placebo or a standardised Bacopa monnieri extract (150 mg twice daily). Outcome measures included the Bergen Insomnia Scale (primary outcome measure), Functional Outcomes of Sleep Questionnaire, Pittsburgh Sleep Diary, Short Form-36 Health Survey, and the Depression, Anxiety, and Stress Scale. Changes in salivary concentrations of cortisol, dehydroepiandrosterone sulfate, immunoglobulin A (sIgA), α-amylase (sAA), C-reactive protein, melatonin, and the fatigue biomarker index were also assessed. Based on the Bergen Insomnia Scale, Bacopa monnieri did not improve sleep patterns more than the placebo; however, it was associated with greater improvements in emotional wellbeing, general health, and pain-related symptoms. Bacopa monnieri was also associated with greater reductions in sIgA and sAA compared to the placebo. Future clinical trials using varying doses, treatment periods, and objective outcome measures will be important to validate these findings

Examples of the management of bacterial zoonoses in Europe and across the globe demonstrate that One Health approaches of international surveillance, information-sharing and appropriate intervention methods are required to successfully prevent and control disease outbreaks in both endemic and non-endemic regions.
